Bryan Johnson: I Just Took the Most Powerful Dose of DMT in the World... Here's What It Was Like

Relinquishing “all attachment, all preconditions, all want, all desire” unlocked what he called his most dynamic human experience, followed by childlike excitement, humor and less defensive conflict with his partner.
The downside is governance-grade, not merely clinical: Friedberg raised permanent psychosis, schizophrenia risk, bad trips and founders abandoning companies or families after radical value changes. Johnson acknowledged that investors have inserted no-psychedelics clauses into deal documents; quantified dosing, licensed supervision and proper set and setting might reduce risk, but “some people may simply not be appropriate candidates.”
The conversation raised whether rapid psychological rewiring could flip from liability to asset as AI accelerates the pace of change. Friedberg suggested psychedelics might help humans adapt, while Johnson described profound “psychoflexibility”; the same mechanism carries tail risk that identity, priorities and obligations change. Johnson reported returning more committed to his work and fatherhood.
The longevity pipeline extends well beyond psychedelics into mitochondrial augmentation, FOXO3 delivery, personalized organoids and computational cell reprogramming. Johnson plans an exploratory mitochondrial procedure, wants to replicate a reported FOXO3 tissue-regeneration result, and has created heart, liver and lung organoids from his induced pluripotent stem cells—although testing has not begun and he repeatedly marked the concepts as TBD.
Johnson sees GLP-1s as longevity’s first big “drop,” with NewLimit or plasma-based FOXO3-like therapies potentially providing a later visible proof point. The bottleneck is control: Friedberg warned that overdosing even one cell with Yamanaka factors could initiate a tumor and theorized that feedback-controlled cellular switches would be needed.

1. 5-MeO delivered a subjective reset within seconds
Two days after the experiment, Johnson said 5-MeO-DMT—“somewhere between 5 and 10 times more powerful than DMT” in his framing—had left him “stunned” and “speechless.” He received 9 mg intramuscularly plus 7.18 mg vaporized; within about 10 seconds, “you’re just out.”
Unlike visually rich DMT experiences, this had “very, very little visual” content. Johnson instead described confronting “raw consciousness and raw intelligence” at an effectively unbounded scale, with the fear that existence might “tear you to shreds” or threaten his sanity.
The decisive fork was resistance versus surrender: he said he could either wrestle with the experience and wait it out or relent by releasing self, ego, desire and every precondition. Full surrender opened “unimaginable bliss and euphoria”—an experience he ranked above accomplishments, marriage, childbirth and overcoming hardship.
The aftereffects were mundane but striking to him: laughing during a dream, waking with childlike anticipation, making immediate quips and resolving conflict with his partner Kate without defending, escalating or storing ammunition for later. He nevertheless asked the load-bearing questions: “How long will it last? What’s the decay curve like?” and will the former Bryan return in 30 or 60 days?
2. The longevity thesis rests on suggestive psilocybin signals
Johnson’s five-year program initially ranked interventions by published evidence and effect size: exercise, nutrition and sleep first, then sauna, hyperbaric oxygen, rapamycin and metformin. Psychedelics entered only after his team saw preclinical psilocybin evidence in mice, including lower inflammation—a recurring feature of longevity therapies.
He then completed three 25 mg psilocybin doses while Kernel measured brain activity. Johnson’s airport analogy: ordinary brains maintain heavily traveled routes such as New York–London; psilocybin effectively “picks up” the airports and rearranges traffic, quieting established connections and permitting new ones. Friedberg supplied the interpretation—new connections can enable new behaviors—and Johnson replied, “Exactly right.”
Johnson reported a metabolic reset: blood glucose moved from the top 99.5% of the population to the top 99.9%, his microbiome changed, and he said Kernel showed dramatic restoration of youthful brain patterns. For 5-MeO, however, the honest result was “so far, nothing”: structural MRI, functional MRI, optical Kernel measurements and real-time EEG had been collected but not analyzed.
3. Clinical and founder risks survive better set and setting
Friedberg’s clinical pushback was explicit: psychedelics can leave some users scarred, precipitate permanent psychosis or push predisposed people toward schizophrenia. Johnson agreed that this was “not a nonzero percentage” risk and that bad trips could leave people scarred; Friedberg said the compounds deserve “all the caution in the world.”
Johnson argued that casual mushroom use often combines unknown strains, unknown psilocybin doses, poor set and setting, and no supervision. Quantification, licensed professionals, appropriate set and setting, and excluding unsuitable candidates could create a safer structure, but he emphasized that much of the risk persists.
Friedberg then moved from patient risk to counterparty risk: he described stories of CEOs emerging from psychedelic experiences, abandoning companies and families, and leaving investors and employees “high and dry.” Johnson had heard of investors contractually barring founders from psychedelics for the company’s duration; his own result was the opposite—greater motivation, with no desire to disappear into the woods.
The deeper issue was whether a neuronally rewired person remains the same person, especially when values and obligations can change. Johnson said fatherhood remained central, but accepted Friedberg’s distinction between support and enablement. Asked about neural implants, transgenic plasmids and his ultimate experimentation limit, Friedberg answered “Yes” to both technologies and “No” to having a limit.
4. The next longevity stack moves into cells, genes and organoids
Having “knocked out” the interventions available today, Johnson is waiting on cell and gene therapies. His nearest experiment is mitochondrial augmentation: a third party will draw blood, expand mitochondria from a biological match and return them in an exploratory procedure. Johnson said he would have the blood draw within a week or two; Friedberg said the therapy was in phase 2.
Friedberg suggested intramuscular delivery and sprint performance as a functional readout, which Johnson immediately called a great idea.
Both discussed a reported FOXO3 delivery system using mesenchymal stem cells, which Johnson said showed rejuvenation across more than 50% of tissues. He contacted the Chinese professor associated with the work and wants either replication or an in-house build—nominally a two-year project, though both thought AI could now compress the setup.
Johnson has also converted his cells into induced pluripotent stem cells and derived heart, liver and lung organoids. The aim is to test molecules, combinations and doses in a dish before exposing his body, gaining time and screening for complications. Johnson said no first test had run; Friedberg called the concept compelling but said it was still TBD whether it works.
As an investor in NewLimit, Johnson said computational discovery of reprogramming factors is advancing faster than expected. Friedberg identified dosing as the existential constraint: one overdosed cell could become a cancer cell and take off as a tumor. He theorized that intracellular feedback switches measuring gene expression and turning protein synthesis on or off could solve the problem; Johnson agreed.
5. Psychological youth may be psychedelics’ differentiated use case
Johnson said psychedelics may not be meaningful for basic bodily functions. Their niche may be restoring a youthful “disposition toward reality”: ambition, curiosity and the feeling that anything is possible. He described 5-MeO as “30 or 40 years of psychological rejuvenation”—a snapback that sleep, diet and sauna never produced.
Friedberg raised whether humans can change fast enough in an AI-accelerated world and suggested psychedelics might become an ally for adaptation. Johnson described Friedberg’s openness to such change as “profound psychoflexibility”; the discussion retained the tail risk that personality and priorities could move with it.
Johnson separately argued that abundance in food, energy, housing, mobility or life improves collective wellbeing. Friedberg then cited 84% of people having metabolic disorders and more than 40% being obese, alongside poor sleep, phone dependence and mental-health issues. Better health, in Friedberg’s view, could restore a collective “can-do attitude.”
